Comparison of the impact of allelic polymorphisms in PfAMA1 on the induction of T Cell responses in high and low malaria endemic communities in Ghana

Abstract Background Malaria eradication requires a combined effort involving all available control tools, and these efforts would be complemented by an effective vaccine. The antigen targets of immune responses may show polymorphisms that can undermine their recognition effectors hence render vaccines based on antigens from single parasite variant ineffective against other variants. This study compared the influence allelic in Plasmodium falciparum apical membrane 1 ( Pf AMA1) peptide sequences three strains P. (3D7, 7G8 FVO) function as immunodominant T cell high low malaria transmission communities Ghana. Methods Peripheral blood mononuclear cells (PBMCs) 10 subjects area (Obom) (Legon) were tested 15 predicted CD8 + minimal epitopes within AMA1 multiple using IFN-? ELISpot assay. peptides also similar assays enriched PBMC fractions same to characterize responding subsets. Results In unfractionated PBMCs, two area, Obom, responded positively four (26.7%) peptides. None Legon subject PBMCs yielded positive PBMCs. with Obom made recall six (40%) peptides, while only one response peptide. Overall, 5 20 who had peptide-specific Obom. Furthermore, strains, 3D7 strain only. Conclusions current data demonstrate possibility real effect induction exposed subjects, this more pronounced higher exposure.
